The invention relates to a method for the determination of an analyte in a sample by fluorescence correlation spectroscopy, the distance between the measurement volume in the sample and the optical excitation/detection direction being .gtoreq.1 mm and the sample liquid being thermally insulated from the optical instrument. The method is especially suitable for the measurement of temperature-dependent processes, for example the determination of nucleic acid hybridization melting curves and/or for carrying out nucleic acid amplification reactions. A device suitable for carrying out the method according to the invention is furthermore disclosed.
