Provided are isolated antibodies or fragments thereof which bind a peptide consisting of residues Ala.sub.22-Glu.sub.23-Gly.sub.24-Gln.sub.25-Leu.sub.26-Gln.sub.27-Trp.sub-.28-Leu.sub.29-Asn.sub.30-Arg.sub.31-Arg.sub.32-Ala.sub.33-Asn.sub.34-Ala.-sub.35-Leu.sub.36-Leu.sub.37-Ala.sub.38-Asn.sub.39-Gly.sub.40 (peptide 307) of mature human TNF-.alpha.. The antibodies and fragments thereof may be used to identify antibodies capable of binding in the region of mature human TNF-.alpha. of amino acid resides 22-40. Antibodies or fragments thereof which bind particular regions of mature human TNF-.alpha. are shown to elicit particular biological activities dependent upon the particular region wherein binding occurs.
