Abstract
The invention relates to a T-cell activating bispecific antigen-binding molecule containing the first and second antigen-binding fragments, one of which is a Fab molecule having the ability to specifically bind to the T-cell activating antigen, and the other is a molecule Fab having the ability to specifically bind to the target cell antigen, and the Fc domain consisting of the first and second subunits having the ability to stable association; wherein the first antigen-binding fragment is a cross-linked Fab molecule, in which either the variable or constant regions of the light chain Fab and the heavy chain Fab are exchanged, wherein the first and second antigen-binding fragments are fused together; and wherein the T-cell activating bispecific antigen-binding molecule contains no more than one antigen-binding fragment having the ability to specifically bind to the T-cell activating antigen. The invention also relates to a isolated polynucleotide encoding a T-cell activating biospecific antigen-binding molecule, vector, host cell, method for producing this molecule, pharmaceutical composition, use for the preparation of a medicament for the treatment of cancer in an individual, and a method for inducing lysis using a T-cell activating bispecific antigen-binding molecule.
